New research lists cell stiffness as possible biomarker

New Georgia Tech research shows that cell stiffness could be a valuable clue for doctors as they search for and treat cancerous cells before they're able to spread. The findings, which are published in the journal PLoS One, found that highly metastatic ovarian cancer cells are several times softer than less metastatic ovarian cancer cells.

Assistant Professor Todd Sulchek and Ph.D. student Wenwei Xu used a process called atomic force microscopy (AFM) to study the mechanical properties of various ovarian cell lines. A soft mechanical probe "tapped" healthy, malignant and metastatic ovarian cells to measure their stiffness.

"In order to spread, metastatic cells must push themselves into the bloodstream. As a result, they must be highly deformable and softer," said Sulchek, a faculty member in the George W. Woodruff School of Mechanical Engineering. "Our results indicate that cell stiffness may be a useful biomarker to evaluate the relative metastatic potential of ovarian and perhaps other types of cancer cells."

Just as previous studies on other types of epithelial cancers have indicated, Sulchek also found that cancerous ovarian cells are generally softer and display lower intrinsic variability in cell stiffnesss than non-malignant cells.

Sulchek's lab partnered with the molecular cancer lab of Biology Professor John McDonald, who is also director of Georgia Tech's newly established Integrated Cancer Research Center.

"This is a good example of the kinds of discoveries that only come about by integrating skills and knowledge from traditionally diverse fields such as molecular biology and bioengineering," said McDonald. "Although there are a number of developing methodologies to identify circulating cancer cells in the blood and other body fluids, this technology offers the added potential to rapidly determine if these cells are highly metastatic or relatively benign."

Sulchek and McDonald believe that, when further developed, this technology could offer a huge advantage to clinicians in the design of optimal chemotherapies, not only for ovarian cancer patients but also for patients of other types of cancer.

This Sunday, my family and I will be participating in the Ron Santo Walk to cure juvenile diabetes 2012 on the Chicago lakefront. We have done this for the past seven years ? ever since we moved back to Chicago from Springfield. My daughter, Grace, developed type I diabetes just after she had turned 2. In fact, I was already in Springfield doing my fellowship in hand and microsurgery, when I came back to visit the family for a weekend in preparation for their move down to the Land of Lincoln. They were packed and ready to go the next week down to Springfield. It was all going to be a great adventure, and we were all very excited about the prospects. But the weekend when I came down, Grace was not feeling so well, and so we took her to the doctor, and a urine dipstick analysis revealed that she had a large amount of sugar in her urine. She had diabetes, and my wife and I were devastated.

We took her to Children?s Memorial Hospital where she was admitted and stayed for about a week. She eventually recovered, and we did go down to Springfield. It was not a particularly easy year in many regards. We were far from our families. I was working very hard, and my wife had to put her residency on hold in order to take care of Grace. But in spite of these difficulties and challenges, we were able to survive and ultimately thrive as a family, and really, before we knew it, I was done with my fellowship in hand and microsurgery, and we were ready to go back to Chicago!

And in the fall of that year, we did our first JDRF walk. And ever since that time we have looked forward to this event because it?s really a wonderful event a beautiful place for meaningful and important cause ? especially in my life.

So today, I wanted to share an interview that I did with my daughter, Grace, and let her talk a little bit about what is like to have type I diabetes. I hope you find it both interesting and informative.

In the next portion of the podcast I talk about the controversy that has been occurring with Omega-3 fish oils ? specifically that recent studies have suggested that they are largely ineffective as supplements. I?ve looked at the study, and of course, it?s one of those observational studies that looks at one particular thing ? taking Omega-3 fish oils ? without necessarily looking at subgroups of people who take them, specifically those who are still eating tons of carbohydrates in the form of wheat and sugar, as well as those who were still eating Omega-6 oils in the form of industrial seed oil. In this segment, I talk about looking at the big picture.

In the final segment of the podcast I answer a question that I have been posed several times in the course of writing my blogs and podcasts, etc. which is: how people react in the medical community might tell about low carbohydrate diets? Well, the fact of the matter is I don?t tell them because many that I have told are skeptical. They really honestly believe that a low-fat, low cholesterol, high carbohydrate, high healthy whole grain diet is the way to go, when in fact, it seems for the majority of people, this is not only a diet that doesn?t help you lose weight but one that encourages weight gain over the long term.

So, I don?t necessarily waste much time trying to convince doctors who have been practicing for 40 years under the low-fat paradigm. That?s one of the funny things about ?doing something.? Whenever you do something, whether it works or not, if you are personally successful at it you feel like you are an expert at some level. And that?s unfortunate because you don?t necessarily leave your mind open to alternate/competing hypotheses. And for some, this expertise often becomes a privileged viewpoint where a principled objection is not seriously considered. It?s actually not a privilege viewpoint ? it?s a blind spot.

That?s why I always try to repeat the fact that as a doctor, I am always first, a teacher ? that is the Latin root of the word! And as a teacher, I am always a continual learner because the best teachers do continually learn. And who do they learn from? Well there?s no doubt that books are important, because they help codify knowledge and principles, but I feel that the central source of learning in terms of medicine comes from people, specifically patients.

Patients of taught me so much, and many times they have completely contradicted what I?ve learned in standard books. As a result of contradictory evidence and information that I have gotten from direct patient contact, I had to go back to books to see why this is so. The reason is simply because human metabolism is very dynamic, and although some people can thrive in a low-fat diet, most do better on a low carbohydrate high-fat diet.

To be clear, there is no one-size-fits-all diet. And the great thing about people who believe in low-carb, like me, is that we are the first to admit it in no uncertain terms. I believe in individualization based on the specific needs of a given patient ? most doctors do? in theory. But when it comes down to the nitty-gritty of crumpling up some of their conventional wisdom and tossing it in the intellectual wastebasket, that is a different matter altogether the most doctors are not inclined to do.

And that is why I don?t necessarily spend a lot of time trying to convince people about a low carbohydrate high-fat diet because in order to do this, I have to first become a psychologist! I have to explain to them why they?re thinking may be wrong and why they?re relying a bad thought process which is ultimately resulting in bad decisions. But nobody likes this. Especially any expert who believes they are functioning at a privileged position of knowledge. They don?t like to be told that what they are confusing as a privileged position of knowledge is actually a blind spot.

So this is why I focus on patients, and I try to achieve objective results treating patients. This is the most important thing as a doctor.

In suitcase at LAX: Smoke grenade, leg irons ...

A 28-year-old man travelling from Japan to Boston was arrested at Los Angeles International Airport after investigators found him wearing a bulletproof vest and carrying a suitcase containing a smoke grenade, a biohazard suit, a gas mask and leg irons. TODAY's Natalie Morales reports.

A Massachusetts man has been charged with illegally transporting a smoke grenade in his checked suitcase while returning from a trip to Japan.

Agents from Immigration and Customs Enforcement arrested Yongda Huang Harris, 28, a naturalized U.S. citizen of Chinese descent, Friday in Los Angeles. He was flying from Japan, through South Korea and then Los Angeles, on his way to Boston.

What alerted agents to check his suitcase? ?It might have been the bullet-proof vest or the flame-retardant pants that he was wearing under his trench coat.

According to ICE, a search of his checked bag turned up the smoke grenade as well as "three leather-coated billy clubs, a collapsible baton, a full-face respirator, various knives, a hatchet, body bags, a biohazard suit, handcuffs, leg irons, and a device to repel dogs."

He appeared late Tuesday in federal court in Los Angeles. There's no thought he was a terrorist or was plotting to do anything on the various planes he was flying on, a federal official says. But transporting a smoke grenade is illegal.

ICE isn't saying what airline allowed him to begin his journey. Court documents say his trip originated in Kansai, Japan, stopped over in Inchon, Korea, and then in Los Angeles on his way to Boston. ?A federal official says the Japan to Korea flight was not on a U.S. carrier.

Russian nationalists want wedding shooters jailed

MOSCOW (AP) ? It was an unusual wedding escort even for Moscow's brash style: A red Ferrari led a motorcade in which guests fired celebratory shots from car windows as they sped down one of the city's main avenues near Red Square.

Sunday's parade drew an angry reaction from the Kremlin-controlled parliament, where senior lawmakers voiced outrage Monday after wedding guests from the province of Dagestan in Russia's North Caucasus walked away with $3 fines. Just one man was ordered to pay a $60 fine by police who stopped the motorcade just outside the Kremlin.

Video of the incident also prompted angry comments from ethnic Russians who denounced the wedding traditions brought into the Russian capital from the volatile Caucasus. Some called for tougher punishment for such behavior.

Vladimir Zhirinovsky, leader of Liberal Democratic Party, said Monday while people from the Caucasus see shooting guns as "a sign of joy," "here it's a sign of robbery, banditry." He suggested such incidents be punishable by up to two years in jail.

Sergei Zheleznyak of the pro-Kremlin United Russia party suggested that celebratory gunfire should land a 15-day jail sentence and substantial fines.

"Meager fines for hooliganism with the use of firearms are a mockery of law and common sense," he said.

Millions of mostly Muslim natives of Dagestan and other Caucasus provinces flooded into western Russia after the 1991 Soviet collapse. Some have formed ethnic gangs notorious for their cruelty, and many others have been involved in violent clashes with ethnic Russian nationalists and soccer fans.

Many Russians harbor anti-Caucasus sentiments, and even those who would not describe themselves as racist are resentful of the hefty subsidies sent to the Caucasus, particularly to Chechnya.

The money is designed to bring stability after two separatist wars in Chechnya, but the region remains deeply impoverished while provincial leaders and officials flaunt their wealth and are often seen driving expensive cars and throwing extravagant wedding parties.

A 19-year-old Chechen man was arrested for three days and fined $150 in 2010 after drunk-driving his SUV over the Grave of an Unknown Soldier just outside the Kremlin.

Alexei Navalny, an anti-corruption lawyer and popular blogger who has helped organize a wave of massive anti-Kremlin protests, said the wedding guests got away with tiny fines because of their connections.

"This single episode completely changes our ideas about what hooliganism and anti-social behavior are," he commented in his blog Monday.

Boutique owner bets on Hertel Avenue - Buffalo Rising

Annemarie Schneider is not averse to risk. ?August 1st, 2011 she opened Chic and Sweet, a women's clothing boutique in North Buffalo, betting on the emerging commercial vitality of Hertel Avenue. ?So far so good. ?

The store's sales have grown steadily and Schneider is now also offering a selection of clothes and accessories online. ?Using Facebook and other social media sites, she has generated sales from all over the country. ? She targets women looking for clothing with classic design elements that are trendy at the same time. ?Her clients range in age from 25-75.?

Schneider has expert advice on marketing to this customer base. ?Her business partner - and husband - Zack Schneider, is also co-owner of 15 Fingers LLC, a marketing agency that specializes in catalyzing audience engagement with online and traditional marketing tools.

And as if all of this wasn't already exhausting, the couple had their first child, August Peter seven months ago. ?Fortunately, they have a large extended family that is only too happy to assist with child care. ?And Augie regularly accompanies his mother to the store, much to the delight of shoppers. ?

Schneider and her husband were eager to start a retail business on Hertel but first patiently conducted research. ?Initially they considered opening a lingerie boutique. ?But when the economy failed to rebound as predicted and consumers pulled back on purchasing luxury items, they decided instead to pursue their current product line.?

Retail can be very challenging under any circumstance because of the cost of inventory. ?Schneider's strategy has served her well since launching her business. ?She views much of her clothing and accessories as "transitional" in that they can be sold and worn during more than one season. ?In addition, she keeps her order quantities low, buying only one or two garments in each size. ?This has an added benefit, according to Schneider, because her clients know they are buying something unique. ?"We pride ourselves on giving our customers an edge on fashion. ?They won't see themselves coming and going when they shop at Chic and Sweet."

Schneider is not new to entrepreneurship and the demanding schedule needed to make a retail venture successful. ? She has always been "obsessed" with fashion design and, after high school, created her own line of jewelry which she sold at home parties. ?She also worked in retail while going to school and gained valuable skills in business management and sales. ? ??

It was during this time that Schneider also attended Canisius College and earned a dual degree in English and Communications which led her to a job at Buffalo Spree Publishing before pursuing her current career in teaching. ?This fall, she started her fourth year as a Reading Specialist in the Lancaster School District. ?Her hectic schedule still allows her to closely manage Chic and Sweet and their four part-time employees. ?Her business is clearly a passion and its success is a priority.

Schneider is very excited by the recent commercial growth on Hertel. ?The opening of Spot Coffee and Globe Market represents significant investment and has increased foot traffic. ?New restaurants such as Mes Que and The Village Beer Merchant are also indications of other entrepreneurs' confidence in the future of Hertel Avenue. ?

While there are key geographic and demographic differences, Hertel is often compared to the Elmwood Avenue strip. ?Hertel may be approaching a critical mass of shopping venues and restaurants, but Schneider feels that a united marketing effort, similar to that of the Elmwood Village Association, is needed if the street is to draw a comparable volume of traffic.?

To that end, she has been meeting routinely with other store owners to discuss ways to collectively promote Hertel from Parker to Delaware. ?For example, more year-round events, such as the Hertel Holiday Walk in December or Shop Rock and Stroll Friday nights during the summer, could bring many new shoppers to the area.?

If their venture continues to thrive, Schneider and her husband may consider opening another business. ?"We really love the city and want it to be vibrant. ?We are always thinking bigger and better." ?Hopefully, they'll take another chance on Hertel. ?

Rallying the troops: Floyd County Republicans bring five big-ticket speakers to rally

NEW ALBANY ? Republican leaders locally and across the state shared a common message at the Republicans of Floyd County?s Reagan Day Rally ? they have 52 days to energize their base to take back Congress and the executive branch.

Five leaders spoke to about 50 supporters Saturday morning about what they thought would help get the country and the state back on track.

Richard Mourdock, the state?s treasurer and republican candidate for U.S. Senate against Democrat Joe Donnelly and Libertarian Andrew Horning, said in terms of the presidential race, he thought President Barack Obama?s widely criticized ?you didn?t build that? comment shows that he?s out of touch with the country and his ideas about increasing tax rates on the highest earners in the country.

?That statement told me, too, that Barack Obama doesn?t understand America and he doesn?t appreciate the potential of capitalism,? Mourdock said. ?It?s not an evil force.?

He said this November?s election is a choice of two visions for the county ? one of American exceptionalism and another of what he described as the European model that demands more of citizens as government grows.

But he said he thinks the Republican Party has a ?secret weapon? in its ability to mobilize its supporters like it did in 2010 and in the May primary election.

Todd Young, who is seeking re-election to Indiana?s 9th district in the House of Representatives against challenger Democrat Shelli Yoder, said he?s optimistic for America?s future, but not without some concerns.

?Now when I look into the future, however, I pause,? Young said. ?I?m not pessimistic because I think we have the ability to do the very same thing Americans have always done. I pause because there are two visions competing for the future of this country.?

He said he thinks the Republican agenda puts its trust in the American people while Obama?s holds the private sector and private individuals responsible for some of its economic woes.

But he also criticized the president for his stimulus package from 2009, as well as his approach to health care reform, government regulations on business and support of tax increases on ?the very people that will get us out of this mess.?

But he said he still has hope for the country.

?You cannot staunch the spirit of the American people,? Young said. ?I still believe in the American people, I know you still believe in the American people. These stories I?ve told today can either define America?s future as they have defined America?s past, or they can become long-lost relics of a once-great nation.?

Tony Bennett, who is seeking re-election as the State Superintendent of Public Instruction and will face Democrat Glenda Ritz, said his approach to education reform has garnered a lot of negative attention, but he thinks the results have proven themselves.

He said? with 85.7 percent of Indiana high schoolers graduating and an 8 percent overall rise in state test scores, those two things show what the changes to the public school system have done.

?Some of you probably go to Little League parks, go to church or social events with teachers and you?ve heard this line: ?Tony Bennett is out to destroy public education ??? Bennett said. ?If that was what we ran on and what we tried to do for four years, we have failed.?

He also said the work of Rhonda Rhoads, Indiana?s representative for the state?s 70th district and Ron Grooms, Indiana State senator for the 46th district, helped set the agenda for education reform in the state, but faced tough opposition.

?The toughest thing they had to do was go home and face stacked town hall meetings by the teachers unions and have a lot of vitriolic rhetoric spewed their way,? Bennett said. ?And they?ve stood in the face of that, they continue to stand in the face of that, and I commend [them] for that.?

He said he thinks his opposition doesn?t have much of a plan other than to reverse what his team has done and charged conservatives to challenge his opposition on that.

Dave Matthews, Floyd County Commissioner, spent a lot of time comparing some of Ronald Reagan?s speeches from 1964 to the times he feels the country is facing now. He said this year?s election marks an important part of the country?s future.

?This year, we stand at the doors of a great decision,? Matthews said. ?We can choose to continue to follow the policies of enslavery [sic] to a failing economy, and increasing government control of our lives, or we can choose to follow the only paths that have ever brought American greatness.?

Matthews faces Democratic incumbent Chuck Freiberger in November.

Last to the podium was Mike Pence, Republican candidate for the governor?s seat. He noted that the optimism of Indiana residents seems to be down when it comes to government and what it does for them.
